Έξι μήνες έχουν περάσει από τη διάσωση του μικρούλη Νέμο. Η ηρωίδα αυτής της περιπέτειας, η αμνησιακή Ντόρι, ζει ήσυχα ανάμεσα στα άλλα ψαράκια κλόουν. Σε μια επίσκεψη του σχολείου της πλάι στον Νέμο για να δουν ψάρια που αποδημούν, αισθάνεται νοσταλγία για το δικό της σπίτι και μαζί της έρχονται παιδικές μνήμες. Θυμάται κάτι για ένα κόσμημα του Μοντερέι στην Καλιφόρνια. Μαζί με τον Νέμο και τον Μάρλιν, ξεκινάει το δύσκολο ταξίδι για να βρει την οικογένεια της και μαζί καταλήγουν στο ινστιτούτο θαλάσσιας ζωής της ακτής Μοντερέι, όπου συναντούν τον Μπέιλι, μια λευκή φάλαινα, την Ντέστινι, μια φάλαινα καρχαρία και το χταπόδι Χανκ, οι οποίοι γίνονται οι οδηγοί της Ντόρι.
